I've only been to Venice, Rome and Barcelona but they are fantastic! I am so jealous you get to see carnival, I obviously wasn't there at the right time but I saw all of the postcards and bought some mask earrings, no matter what you've read you won't be prepared for this suprisingly dumpy, marvelous place! You can see all of the sites in Rome in 3 hours if you have a good sense of direction and don't dawdle, but I say you should dawdle and take it all in. In Barcelona, PARTY.
